Depression, variously called Melancholia, Depressive reaction or Neurosis and major depressive disorder, has been a major cause of disability and suicide through history, and is the most common mental disorder and the leading psychiatric complaint prompting medical consultation in most series.

CLINICAL FEATURES / SYMPTOMS OF DEPRESSION


The manifestations of major depressive disorder are multiple and pervasive, and its effect on life function and quality of life is comparable to that of diabetes.
The chief characteristic of a major depressive episode is : low mood, with anhedonia or inability to derive pleasure from formerly enjoyable activities and pursuits.
  • Rumination over feelings and thoughts of regret, hopelessness, helplessness and unworthiness are common, as are preoccupation with guilt and self-deprecation.
  • Depressive symptoms may be accompanied by symptoms of psychosis, particularly self-deprecating or –accusatory delusions or less common frightening or menacing hallucinations.
  • Psychotic depressive episodes or major depression of the melancholic type are often associated with impaired concentration and memory, while social withdrawal, abandonment of former activities, decreased sex drive and interest and preoccupation with death or thoughts of suicide are common.
  • Sleep disturbance is almost universal, usually early awakening with inability to get back to sleep but also hypersomnolence.
  • Multiple physical complaints, chiefly digestive ones or related to headache or fatigue, are common, and in developing countries these are the most common presenting symptoms of depression.
  • Decreased appetite and weight loss are more common than increased appetite and weight gain, and recent cognitive symptoms such as forgetfulness and motor symptoms such as slowness are described, particularly in older depressed individuals.
  • Either agitated or lethargic behavior may be reported by caregivers of older patients, in whom coexistent physical disorders such as cardiovascular disease, stroke or Parkinson’s disease are more common.
Quote:Mood may be irritable rather than depressed in children, and personality change, inattention and decreased cognitive performance in school may predominate over depressive feelings.

What is Colorectal Cancer?
It is a form of Cancer that affects the 'Colon' and/ or Rectal regions of the large intestine. In severe form, following is how it may appear:

What are the colon and rectal regions of the intestine? | Understanding Colorectal Cancer
Colon is commonly known as 'Large Intestine' as well, though both colon and rectum are part of Large Intestine. Colon is the region where water is absorbed and remaining waste material is stored as feces. In the typical diagram of the digestive system, Large intestine (or colon) surrounds the longer (small) intestine.
Rectum is the final section of the large intestine just before anal region where feces are temporarily stored. Following image should explain it all:

What are the Symptoms of Colorectal Cancer? | Understanding Colorectal Cancer


It often starts with change in the bowel habits i.e :
a) Rectal bleeding while passing feces
b) In-consistent and narrowed stools (due to partial intestinal blockage by the tumor)
c) Blackish bowel movements (also called Melena)
d) Mucus in the stool due to excess discharge by the epithelial cells
e) Persistent diarrhea or constipation
f) Feeling a filled gut despite passing the stools
h) Frequent need to pass stools especially 'immediately after meals'
i) Problem of persistent gas/ bloating/ fullness and cramps in abdomen  
j) Feeling a lump in abdomen
k) Feeling of nausea/ vomiting/ loss of appetite/ fatigue
l) Weight loss and anaemia due to blood loss in abdomen
m) Frequent urinary tract infection due to perforation of the epithelium/ inner lining of abdomen
 
Quote:Cancer in ascending colon usually results in Black, tarry bowel movements, called Melena.
While cancerous growths in descending (left-side) colon or rectum result in rectal bleeding (blood mixed or coated on stools).

Advanced Colorectal cancer can be characterized by its spread to other organs of the body leading to some or all of the following symptoms:
Abdominal Pain that can't be tolerated
  • Ascites i.e build of fluids in abdomen
  • Jaundice
  • Hepatomegaly or enlarged Liver
  • Hip/ buttock pains
  • Enlarged lymph nodes
  • Difficulty in Breathing
  • Appetite loss

I write as one with a B.S. in Biology but from 20 years ago.  Have recently taken interest in gene engineering.  Very much would like to get up to speed with gene engineering as one might find at the website DNA 2.0

There is the commercial side at: https://www.dna20.com/services/gene-synthesis  and some learning that is, apparently, for the already skilled. Perfusion Cultures

Hello, my name is Arthur. 

I am willing to explore perfusion bioreactor cultivation specifics. Almost all of the information I have gathered over the past 2 months are articles, in which researchers use perfusion bioreactor setups for cultivating mammalian cells. I am willing to perform a similar experiment using TFF, but there is a problem, considering that all mammalia cells seem to be very sensitive to contamination. I won`t be able to perform the experiment on mammalian type cultures, due to the lack of required sterility of the lab in which I work.
Is it possible to perform cultivation processes using perfusion on microbial or yeast cultures? I have performed numerous fermentation processes on Escherichia coli and Saccharomyces cerevisiae in a fed-batch mode, but haven`t found any information about weather I can perform perfusion cultivations on these cultures or some other cultures that do not require a very high level of sterility.

If someone has some experience on this topic or some ideas, please help me with this problem. Also, if someone has some advise about performing perfusion cultivations I would gladly read it. Thank you for your time.))
